The role of the binder excipient is to act as a binder to bind powder, granules and other dry ingredients together to give the product the necessary mechanical strength. These excipients can agglomerate powders into granules in a process called granulation, and also affect their properties, such as compaction, drug release, flow, solubility, and strength. Binder excipients are used in the process of drug or drug preparation to improve the disintegration, volume, bioavailability and dissolution of the drug.

Classification

Binder excipients hold the ingredients of the formulation together, while bestow API with propriate volume. For example, to give tablet the essential mechanical strength, binder retains or binds granules, powders, and other dry constituents together.

Commonly used binders are:

Application

  • Capsules, chewable tablets, direct compression, rolling, sustained release.

  • The dry binder in tablets, capsules and granules is used as a film-forming agent in tablet coatings and coatings, and as a film-forming agent in topical sprays. It is widely used in HME process.

Limiting Factors in the Selection of Adhesive Excipients

1. Adhesives and solvent of adhesives are considered for drugs with heat and humidity instability. Choose anhydrous, low drying temperature adhesive and its solvent.

2. Related to mixing time: the longer the mixing time in the system of soft material, the greater the viscosity of soft material, the harder the particles produced.

3. When the amount of auxiliary materials in the prescription accounts for more than 80%, on the premise of not affecting the nature of the main drug, should focus on the characteristics of the auxiliary materials to choose the adhesive. If the use of sucrose as an auxiliary material, its dosage reaches more than 80%, it is necessary to take into account the characteristics of "sucrose in water viscidity becomes stronger", choose non-water solvent to dissolve the adhesive (only soluble in water but not soluble in organic solvent adhesive is not applicable), reduce the viscosity between particles, relatively enhance the viscosity inside the particles.

4. Different solvents and different concentrations of adhesives have different viscosity and granulation effects.
