Pellets are spherical or near-spherical free-flowing particles with a size usually between 500 and 1500 microns. They are usually produced by a granulation process in which a powder mixture consisting of API and excipient particles is aggregated into spherical particles. They can be formulated as an immediate release or sustained release dosage forms, and can also be coated to deliver drugs to specific sites of action in the gastrointestinal tract. Due to the free-flowing properties, the pellets provide a high degree of flexibility for formulation development. They are easy to pack without any difficulties. The spherical shape and low specific surface area of the pellets make the film uniform. Compared with tablets, the pellets eliminate the dose dumping effect, make the blood drug concentration curve smoother, and the drug absorption is more gradual, which further reduces the adverse drug reactions.
