The authors carefully designed an antitumor nanoplatform, denoted as ZLGCR, by integrating glucose oxidase, LA oxidase, and CpG oligodeoxynucleotides into a zeolitic imidazolate framework 8 disguised with erythrocyte membranes. ZLGCR-mediated Glu and LA depletion not only enhances the effectiveness of metabolic therapy but also reverses the immunosuppressive TME, thereby enhancing the therapeutic effect of CpG-mediated anti-tumor immunotherapy. Moreover, when combined with immune checkpoint blockade therapy, the synergistic effect of metabolic therapy and immunotherapy is further enhanced. This engineered anti-tumor nanoplatform will achieve synergistic tumor suppressive outcomes by modulating metabolic and immune responses within the TME.
