CpG ODNs (CpG oligodeoxynucleotides) are synthetic DNA sequence with unmethylated cytosine-guanine pairs that mimics the immunostimulatory properties of bacterial DNA. As TLR9 agonists, they activate immune cells such as B cells and plasma cell dendritic cells, triggering both immediate and long-term immune responses. Their thiophosphate skeleton enhances stability and cellular absorption, making them more durable than natural bacterial DNA.

Class A is a palindromic sequence with a PTO-based CpG and a poly-G tail at the 3' end. Class A CpG ODNs do not activate B cells but stimulate plasmacytoid dendritic cells (pDCs) to secrete interferon-alpha. Modifications can enhance their stability and immunostimulatory effects.
Class B includes fully phosphorothioate-refined linear CpG ODNs, some of the oldest identified. This class stimulates B cells without activating pDCs to release IFN-. Customization generally involves adjusting sequence length and PTO modification for stability and enhanced immunostimulatory effects. Additional modifications may target the CpG motif and backbone to achieve specific immunogenicity titers for therapeutic applications like vaccine design.
Class C includes the structures of classes A and B, with a pure PS backbone and a CpG palindrome motif. It stimulates B cells and pDCs to release interferon-alpha (IFN-), but its effect is less pronounced than that of class A or B ODNs. Sequence modifications can help balance immune activation and reduce toxicity, which is essential for personalizing treatments for diseases like cancer or chronic infections.

Appropriate CpG sequences are selected based on the available literature and databases. Typical CpG sequences consist of unmethylated CpG dinucleotides.

Results:
These CpG-ODNs showed better activity against grouper TLR21s than the currently developed CpG-ODNs and produced similar immunostimulatory effects when applied to cells isolated from orange-spotted grouper. The developed CpG-ODNs were also effective in activating human and mouse TLR9-mediated NF-κB activation and cytokine production. These findings suggest that the GTCGTT matrix is required for activation of grouper TLR21s by CpG-ODNs, and that the CpG-ODNs developed for grouper TLR21s contain structures that effectively activate human and mouse TLR9s.
