What is Peptide Antigen?

Peptide antigens are short chains of 8 to 30 amino acids that can trigger an immune response in the body. These peptide antigens are usually derived from proteins and represent fragments of larger proteins or polypeptides. During the immune response, peptide antigens are received by antigen-presenting cells (such as dendritic cells and macrophages), processed, and presented to T cells in the form of peptide-major histocompatibility complex (MHC) complexes. This process is crucial for identifying foreign pathogens, cancer cells, and other abnormal cells. By designing specific peptide antigens, the body can be induced to produce an immune response against specific pathogens or tumor cells, thereby effectively improving the targetedness and efficiency of treatment.

Design Your Peptide Antigen

Although many peptide sequences may be immunogenic, not all are equally effective in generating antibodies reactive against the target protein. We focus on the following factors during the peptide antigen design process to ensure that you are satisfied with the product you synthesize.

  • Accuracy of the amino acid sequence of the target protein.
  • Predicted higher-order structure of the intact protein.
  • Protein domain/region mimicked by the peptide antigen.
  • Ease of synthesis of certain peptide sequences, ideally, the peptide should be 10 amino acids or more in length.
  • Avoid any cysteine residues whenever possible.

Multi-Antigenic Peptides (MAP)

MAP is a branch of synthetic peptides that have a backbone core of lysine residues to support the formation of a variety of peptide sequences with less than 8 branches. The molecular weight of these peptides (10kDa-15kDa) is ideal because they are highly immunogenic without the need for conjugation to a carrier protein. Our MAP synthesis method is standard SPPS and includes:

  • Anchoring Boc-Lys(Boc)-OH to the resin.
  • Sequential treatment with TFA and deprotection.
  • Coupling and deprotection cycles.

Note that for the extreme C-terminal region of the protein, we recommend the use of peptide-protein conjugates.

Peptide-Carrier Protein Conjugation

Peptide-carrier protein conjugates are another class of peptide antigens that are useful when the epitope is located at the C-terminus of the peptide. In the peptide-protein conjugation process, the peptide is bound to the carrier protein by covalent or non-covalent bonding to form the conjugate peptide.

Technology: Synthesis of Multi-Antigenic Peptides

Results:

The authors describe a method for synthesizing multiple antigenic peptides (MAPs) targeting predicted antigenic determinants of viral antigens. The method involves predicting linear epitopes using a predictive computer algorithm, synthesizing peptides for the predicted regions, and manually synthesizing the MAPs by SPPS and testing them. Eight MAPs were prepared by this method and used to immunize rabbits to generate anti-peptide antibodies. The reactivity of the MAPs in detecting native cognate antigens in whole virus was confirmed by ELISA.
