The authors describe a method for synthesizing multiple antigenic peptides (MAPs) targeting predicted antigenic determinants of viral antigens. The method involves predicting linear epitopes using a predictive computer algorithm, synthesizing peptides for the predicted regions, and manually synthesizing the MAPs by SPPS and testing them. Eight MAPs were prepared by this method and used to immunize rabbits to generate anti-peptide antibodies. The reactivity of the MAPs in detecting native cognate antigens in whole virus was confirmed by ELISA.
