Molecular Weight Determination We determine the exact molecular weight of proteins using high-resolution mass spectrometry (Orbitrap, QToF) and a range of other techniques such as size exclusion chromatography (SEC), and SDS-polyacrylamide gel electrophoresis
Electrophoresis Pattern Analysis We obtain electropherograms and homogeneity and purity data for proteins and peptides using capillary isoelectric focusing (CIEF/iCIEF), polyacrylamide gel electrophoresis, SDS-polyacrylamide gel electrophoresis, and capillary electrophoresis.
Protein Concentration Analysis We use UV/visible absorbance of product solutions of known protein content to accurately determine the extinction coefficient and use amino acid composition analysis to quantify the concentration of protein in the solution.
Isomer Pattern Analysis We use chromatography and gel/capillary electrophoresis for isomer patterning as well as impurity detection.
Protein Aggregation Analysis We perform protein aggregation studies using dynamic light scattering, multi-angle laser light scattering (MALS), and sedimentation velocity analytical ultracentrifugation (SV-AUC).
Chromatographic Analysis We use size exclusion chromatography (SEC), reversed-phase liquid chromatography (RP-HPLC), ion exchange liquid chromatography (IEX), and others to generate chromatograms and obtain data on identity, homogeneity, and purity.
Spectral Analysis We perform protein and peptide structural analysis using far- and near-UV circular dichroism (CD), nuclear magnetic resonance (NMR), infrared (FTIR), or ultraviolet-visible spectroscopy.

Results:

The authors successfully develop an ETD method to facilitate peptide sequence analysis by combining ion/ion chemistry and MS/MS, and use a combination of ion/ion chemistry CAD, and ETD to characterize the primary structure of intact proteins.
