The use of microwave energy to enhance solid-phase peptide synthesis has proven to be a valuable technology. It has been shown to enhance the deprotection and coupling steps in each amino acid cycle. MW-SPPS is a process that uses microwave technology to accelerate solid-phase peptide synthesis. Studies have shown that microwave irradiation significantly enhances amide bond formation in peptide synthesis. This allows for more complete coupling in less time and potentially reduces reagent overuse. At high temperatures, carbodiimide-based coupling is preferred over onium salt-based methods, which use strong bases and lead to increased epimerization.

The authors present an optimized protocol for the rapid synthesis of eptifibatide. Several strategies based on microwave-assisted solid-phase peptide synthesis (MW-SPPS) that are scalable to kilogram-scale manufacturing are investigated. Thiol-free off-resin disulfide bond formation for the production of eptifibatide is compared with four novel and different on-resin approaches, where ring closure is achieved via disulfide bonds and via simple amide bond formation of the MPA head group on the cysteine side chain. The results show that a strategy based on StBu orthogonal protection on selected cysteines can be used in a fully automated solid-phase microwave-assisted cGMP-ready process for the preparation of eptifibatide.
