Microwave-Assisted Solid-Phase Peptide Synthesis (MW-SPPS) Technology

Inquiry

The use of microwave energy to enhance solid-phase peptide synthesis has proven to be a valuable technology. It has been shown to enhance the deprotection and coupling steps in each amino acid cycle. CD Formulation uses advanced microwave-assisted solid-phase peptide synthesis (MW-SPPS) technology to optimize the synthesis of peptides, improving both the efficiency and yield of peptide production and meeting the needs of different customers.

What is Microwave-Assisted Solid-Phase Peptide Synthesis (MW-SPPS) Technology?

MW-SPPS is a process that uses microwave technology to accelerate solid-phase peptide synthesis. Studies have shown that microwave irradiation significantly enhances amide bond formation in peptide synthesis. This allows for more complete coupling in less time and potentially reduces reagent overuse. At high temperatures, carbodiimide-based coupling is preferred over onium salt-based methods, which use strong bases and lead to increased epimerization.

Our Services Related to Microwave-Assisted Solid-Phase Peptide Synthesis (MW-SPPS)

CD Formulation is actively developing the application of microwave technology in peptide manufacturing. Our team often uses this method to process various complex peptides, allowing synthesis in a shorter time and higher yields.

MW-SPSS is a method that uses microwave radiation to increase the efficiency of peptide synthesis. This technology improves SPPS synthesis and can synthesize peptides in high yields with low racemization.

Microwave energy prevents growing peptide chains from folding or forming aggregates and promotes chemical bonding. In other words, peptides can be easily and efficiently stitched together by microwaves, allowing the synthesis of longer peptides and various modifications to the peptide chains, including the addition of sugars and other groups.

Peptide Synthesis and Manufacturing

The steps of peptide synthesis by MW-SPPS are similar to those of SPP, including:

  • Preparation for SPPS.
  • Amino acid addition.
  • Deprotection and cleaning.
  • Chain release.

Scale-up Production

We are equipped with both laboratory-scale and GMP-grade microwave solid-phase synthesis equipment. This equipment can synthesize and manufacture high-quality, competitively priced peptides for research-grade to commercial-grade use.

Our Equipment

  • Automatic microwave solid phase synthesizer.

The authors present an optimized protocol for the rapid synthesis of eptifibatide. Several strategies based on microwave-assisted solid-phase peptide synthesis (MW-SPPS) that are scalable to kilogram-scale manufacturing are investigated. Thiol-free off-resin disulfide bond formation for the production of eptifibatide is compared with four novel and different on-resin approaches, where ring closure is achieved via disulfide bonds and via simple amide bond formation of the MPA head group on the cysteine side chain. The results show that a strategy based on StBu orthogonal protection on selected cysteines can be used in a fully automated solid-phase microwave-assisted cGMP-ready process for the preparation of eptifibatide.
