Peptides are short polymers composed of about 20 ~ 50 amino acids, and protein is a macromolecular and high molecular weight polypeptide composed of more than 50 long-chain amino acids arranged into linear chains by peptide bonds. Therapeutic proteins and peptides have unique and unparalleled pharmacological properties with better therapeutic indices. However, proteins also have the limitation of low bioavailability and are often repeatedly administered through invasive routes, with the advent of oral thin film drug delivery technology, which can significantly improve the biological performance of proteins and peptides. CD Formulation has established an oral thin film protein/peptide technology platform to facilitate the development and investigation of protein/peptide utilizing oral thin film delivery systems, promoting the development of protein/peptide drugs.

CD Formulation is a leader in oral thin film and protein/peptide drug development and manufacturing. Over the past few years, we have been working on applying oral thin film technology to deliver protein/peptide drugs. We have established an advanced oral thin film protein/peptide drug delivery platform to help our customers develop and characterize proteins/peptide drugs that meet their needs.
Oral thin film protein/peptide drugs can comprise one or multiple layers that contain APIs and suitable excipients, such as adhesives to optimize function, and its production methods mainly include solvent casting method, 3D printing method, electrospinning, etc.

The structural stability of proteins or peptides is a major factor in biological activity; therefore, physical and chemical stability must be guaranteed. Our team found that adding excipients such as polymers and sugars used to fabricate oral thin films can confer stability to protein/peptide.
Another innovative delivery platform we are developing to improve the stability of protein/peptide in oral thin film systems is nano- and microparticle technology, which can enhance the permeability of protein/peptide in mucosal epithelial cells, prevent enzymatic degradation and maintain the stability of protein/peptide during processing, manufacturing, and storage.
We provide specialized manufacturing services for oral thin film protein and peptide delivery. This innovative drug delivery system is designed to provide an efficient and convenient method for delivering proteins and peptides through oral administration, overcoming traditional challenges associated with the oral delivery of biologics.

Results: In this study, the author prepared protein-loaded orodispersible films (ODFs) based on blends of trehalose/pullulan by air- and freeze-drying. ODFs were loaded with three model proteins. Trehalose/pullulan-based ODFs are from a technical point promising for possible protein delivery via the oral cavity.
