Results:
In this paper, we successfully established dipeptide nanoparticles/cyclic peptide/aptamer AS1411/influenza hemagglutinin peptide/siRNA/doxorubicin (DNPs/Clolar/AS1411/HA/RNA/DOX) multifunctional nanoparticles for specific delivery, cancer therapy, and bioimaging. It is ideal for the synergistic treatment of multiple drugs. First, the DNPs formed from the self-assembly of dipeptides are non-toxic, providing safe and biocompatible effects. Second, from the perspective of the multi-functional nanoparticles for nano-drug tumors imaging monitoring, AS1411 and HA were used as cell permits for enhancing the specificity of cell drug delivery ability and improving the endosomal escape, respectively. Third, multifunctional nanoparticles synergized with three drugs, Clolar, siRNA and DOX, to improve the therapeutic effect on tumors. Both cellular and in vivo experiments showed that multidrug synergistic therapy was superior to monotherapy. Therefore, the proposal of multifunctional nanoparticles provides new ideas for these hybrid anticancer drugs based on peptide self-assembled nanocarriers and their wide application in biomedicine.
