To guarantee the safety and efficacy of transdermal patches, a thorough assessment of their peel adhesion is necessary. Peel adhesion testing of patches is a crucial procedure intended to replicate real-life conditions and ensure that the adhesive will function optimally during use. CD Formulation is able to offer professional transdermal formulation peel adhesion testing services.
Peeling strength refers to the resistance of the patch's adhesive to being peeled off from the skin. The greater the peeling strength, the more adhesive the patch is. Adhesion is a critical property of a plaster. If the patch has poor peeling adhesion, meaning its adhesive force on the affected area is weak, it may hinder the drug's permeability into the body or cause irritation to nerve endings. Consequently, the medicinal effect may not be exerted effectively, and the patch might even fall off. On the other hand, if the peeling adhesion of the patch is too high, removing the patch from the affected area could leave some adhesive components on the skin. This could result in unbearable pain and might even lead to redness and swelling. Therefore, having an appropriate level of peeling adhesion is essential for ensuring the patch's efficacy and for making it convenient and comfortable for patients to use.
Our steps for transdermal formulation peel adhesion testing are as follows:
Fig.1 Flow chart of transdermal formulation peel adhesion testing (CD Formulation)
1. Place the sample in a laboratory environment at 20°C and 65%RH for 24 hours.
2. Cut 5 specimens with a length of 400 mm and a width of 25 mm from the surface of the sample.
3. Tear off the release paper and stick the sample to the center of the stainless-steel plate so that both sides of the sample are parallel to the two long sides of the steel plate. Use a pressure roller to roll the sample 4 times along the length direction and leave it for 10 minutes.
4. Clamp the free end of the sample and the test plate in the upper and lower clamps of the equipment respectively, so that the long axis of the sample is parallel to the center line of the upper and lower clamps.
5. Set the sample name, sample width, test speed (300 mm/min) and other parameter information, click the test option, and the test starts.
6. During the test, the system displays the force value during the sample peeling process in real time, and automatically displays the test results after the test is completed.
7. Repeat steps 3-6 until all 5 samples have been tested. The test results are the average of the test results of the 5 samples.

Results: In the study, the dosage form characteristics, and technological progress of transdermal patches were reviewed, and the quality control indicators of transdermal patches were summarized systematically. The drug release, transdermal properties, adhesion, peel adhesion, drug residue, and other key quality indexes of transdermal patches were evaluated.
Peel adhesion is an important performance indicator of transdermal patches and an important factor that affects whether the drug can function properly and whether it is convenient to use.
