CD Formulation provides a standardized testing procedure to determine the moisture absorption properties of oral thin film and evaluate its moisture absorption performance in humid environments, providing guidance for the transportation and storage of oral thin film.
The moisture absorption of oral thin film will promote the growth of microorganisms, which will affect the appearance, stability, effectiveness, safety, and other external and internal qualities of the thin film preparation, so it is important to study the moisture absorption of the thin film preparation.

The physical stability of oral thin film at high humidity conditions can be checked by moisture absorption testing. The moisture absorption testing method typically involves saturated sodium chloride method and dynamic vapor sorption (DVS) method.
The moisture absorption testing methods typically involve exposing the sample to a dryer containing a saturated solution of sodium chloride for a period of time. After the exposure period, the sample is weighed to determine the amount of moisture it has absorbed. The percentage of moisture absorption is calculated through the equation formula.
Another more accurate moisture absorption testing method is the DVS method, and our DVS method testing procedure is as follows:

Moisture Absorption Testing | Detailed Information |
Dynamic Vapor Sorption (DVS) | DVS is an advanced moisture adsorption testing equipment, it analyzes the sample's ability to adsorb moisture by the adsorption isotherm of the sample at a specific temperature and pressure. During the testing, the instrument control system automatically adjusts the sample temperature and humidity according to the preset program, thereby achieving dynamic testing of the sample's moisture adsorption performance. |

Results: In this study, montelukast fast-dissolving oral thin films were prepared by solvent casting method with different film-forming agents like HPMC, PVP, PEG 400, glycerol as a plasticizer and mannitol as filler and sweetener. The Percentage moisture absorption (PMA) test was carried out to check the physical stability of films in high humid conditions. In the present study, the moisture absorption capacity of the MTS films was determined by keeping the preweighed films in a desiccator at room temperature for 72 hours. Then they were taken out and exposed to 79.5% relative humidity (saturated solution of aluminum chloride). The results show that the percentage of moisture absorption varied within the range of 2.45 to 5.21%.
