Liposomes are wrapped with particles of various active ingredients, which can efficiently penetrate into the base layer of the skin, and absorb faster and better than ordinary skin care products. When used in basic skin care products, the effectiveness is naturally better than traditional skin care products. Due to its very small diameter, liposomes can easily penetrate the pores and the space between the stratum corneum cells, sending the active ingredients to the dermis and basal layer, allowing the skin to absorb more water and nutrients. If a drug can only penetrate the epidermis but not the dermis, it is referred to as topical administration, which is employed for the treatment of diseases such as skin cancer, psoriasis, eczema, and microbial infections since the root cause resides in the skin. If the drug has access to systemic circulation, it is known as a transdermal drug delivery system (TDDS), highly favored in biomedical fields and suitable for treating various conditions including depression, hypertension, angina, schizophrenia, and Alzheimer's disease. In vitro experiments offer advantages of simplicity, ease of implementation, and result reproducibility; however they fail to reflect effects related to skin blood flow and enzyme metabolism associated with actual cutaneous drug administration. Therefore, further development of transdermal drug delivery systems necessitates in vivo pharmacokinetic studies. In vivo pharmacokinetic experiments primarily investigate laws governing drug absorption,distribution ,metabolism,and excretion within an organism,and play an indispensable role in studying mechanisms of action and bioavailability.
The dose of transdermal administration is generally small, and the drug cannot completely penetrate the skin with a multilayer biofilm structure, so the amount absorbed through the body is small, and the detection of blood drug concentration requires extremely high sensitivity. This is also the biggest challenge in skin testing plasma samples. To meet the requirements of extremely high sensitivity, on the LMS platform, our bioanalysis team comprehensively considers three aspects of mass spectrometry, liquid phase and sample pretreatment to improve the mass spectrum signal response of the compound.
After skin samples are collected in vivo, our in vivo evaluation team has extensive experience in separating layers of skin tissue from frozen sections to meet the testing needs of our customers.
The cuticle is very thin and requires a special sample collection method. Tape stripping sampling method is widely accepted and used, and it is also the FDA recommended cuticle collection method. We have designed a unique set of pre-processing methods, such as ensuring maximum extraction of the analyte. Secondly, considering the cumbersome sample processing process, the working fluid of linear and quality control samples was directly added to the blank tape supernatant containing cuticle for subsequent analysis.

Results: In this study, baicalein-containing nano-liposome gel was successfully prepared by thin film hydration technology. In vitro and in vivo studies were conducted on the enhancement of local permeability of natural flavonoids baicalein by nano-liposome gel, and the formulation was designed and optimized. The effects of independent variables (amount of PC and CHO) on different responses (particle size, encapsulation rate and 12-hour drug release) were studied.
