Glutathione (GSH) is a tripeptide composed of glutamate, cysteine, and glycine, containing a thiol group. GSH exists primarily in two forms: reduced (GSH) and oxidized (GSSG). Functionally, GSH serves as a cofactor for glycerol-3-phosphate dehydrogenase and an auxiliary factor for aldose reductase and glucose-6-phosphate dehydrogenase. It plays a critical role as a cellular regulator and is involved in the tricarboxylic acid cycle and glucose metabolism. Moreover, it can activate various enzymes as a thiol (SH) enzyme cofactor, promoting the metabolism of carbohydrates, fats, and proteins. GSH-liposome formulations offer distinct advantages for applications in cosmetics, pharmaceuticals, and dietary supplements.

In general, these liposomes contain a molecule that reacts with excess glutathione (GSH) in tumor cells, resulting in the depolymerization of the liposomes, the release of the drug, and the depletion of GSH, which weakens the antioxidant capacity of the tumor cells.

Results: In this study, the authors develop maleimide liposome (ML) adjuvants that promote immunogenic cell death (ICD) induction and dendritic cell (DC) maturation by consuming glutathione (GSH) to enhance photothermal immunotherapy for breast cancer. ML effectively depletes intracellular GSH and up-regulates reactive oxygen species (ROS) in tumor cells and DC. In tumor cells, ROS promotes the production of ABTS· +, which activates photothermally induced ICD. In DC, it mitigated immunosuppression and promoted DC maturation (57%) and antigen presentation. With the help of ML, the treatment system increased the infiltration of CD8 + T cells in tumor tissue to 53%, causing a strong distance effect and anti-metastasis effect.
