Immunoprecipitation (IP) is a method based on the principle of antibody-antigen interaction, which is used to specifically enrich and separate proteins of interest and their interacting molecules. Its basic principle is to combine the target protein with a specific antibody to form an immune complex, and then use the affinity of the antibody to the immune complex to precipitate it together with other non-specific proteins. Through steps such as washing and centrifugation, non-specific proteins can be removed, thereby purifying the protein of interest and its interacting molecules.
Co-immunoprecipitation (Co-IP) is a method used to study protein interactions based on the interaction between antibodies and antigens. The principle is to use the endogenous target protein of the cell as bait, and co-incubate the target protein antibody with the total cell protein to promote the formation of immune complexes. Then add protein A/G that can bind to the target protein antibody to form a "binding protein-target protein-target protein antibody" complex. After purification, the immune complex is separated by gel electrophoresis and combined with Western blot or mass spectrometry technology to achieve the purpose of purifying the target protein and qualitatively detecting the antigen or antibody. Compared with other research methods, co-immunoprecipitation detects the interaction between proteins under physiological conditions. Therefore, not only can the complex formed in the body be detected, but also false positives caused by overexpression of the target protein can be eliminated. Second, endogenous target proteins are processed, modified, and mature proteins, so modification-dependent protein interactions can also be detected.
