The following types of drug conjugations can be customized at CD Formulation:

Antibody drug conjugates (ADC) consists of three parts: antibody, linker and small molecule cytotoxin. It uses linkers to couple monoclonal antibodies targeting specific antigens with small molecule cytotoxic drugs. It has both the tumor targeting property of antibody-antigen specific recognition and the powerful killing ability of small molecule drugs. Compared with traditional cancer treatment drugs, ADC drugs not only reduce toxicity by increasing the payload, but also increase the maximum tolerated dose of the drug through their antibody drug properties.

Peptide drug conjugate (PDC) consists of polypeptide-coupled drug, which is composed of linker, homing peptide and cytotoxic payload. Targeting peptide can specifically target overexpressed protein receptors on the surface of tumor cells to deliver cytotoxin and induce tumor cell apoptosis. The drug molecular weight of PDC is smaller, the synthesis and purification of targeted peptides is easier, and the production cost is lower. In addition, PDC drugs have strong tumor penetration and low immunogenicity.

Radionuclide drug conjugates (RDC) is mainly composed of antibodies or small molecules (Ligand), linker (Linker), chelator (Chelator) and cytotoxic/imaging factors (radioisotopes, radioisotopes) that mediate targeting. The biggest difference is the drug load. RDC is no longer a small molecule, but a radionuclide. Using different medical nuclides can enable different functions of imaging or treatment, and some nuclides have both capabilities.

Small molecule-drug conjugates (SMDC): SMDC and ADC have similar composition, both of which are composed of target molecules, linking arms (Linker) and effector molecules (cytotoxicity, E3 ligase). The most direct difference between SMDC and ADC is the targeted molecule. ADC uses biological antibodies as drug targeting, while SMDC uses small molecule targeting. In the preparation of ADCs, the ratio of payload to antibody is often uncertain, and a relatively accurate drug-antibody ratio (DAR) can be achieved by using fixed-point coupling techniques, while the small molecule targeting ligand and payload of SMDC usually have accurate values.

Virus-like drug conjugate (VDC): It is a coupled drug form that utilizes viral capsids designed as non-infectious protein nanoparticles (virus-like particles, VLP) as efficient delivery vectors.

Antibody oligonucleotide conjugate (AOC): It is mainly composed of targeted antibodies, linkers and therapeutic oligonucleotides (siRNA, PMO, etc.), which use antibodies to deliver oligonucleotides to specific cells or tissues, thereby reducing the amount of drug required for treatment and solving the problem of targeted oligonucleotide delivery. AOC drugs can also improve the pharmacokinetic properties of oligonucleotides and expand their application range.
